The stereotaxic instrument, also known as the stereotaxic apparatus, is a three-degree coordinate system specified by landmarks or other reference points outside the skull to determine the location of certain neural structures in the subcortical area for directional stimulation, destruction, injection of drugs, guided potentials, and other investigations under nondirect visual exposure. Stereotaxic instrument are important research devices within the fields of neuroanatomy, neurophysiology, neuropharmacology, and neurosurgery for oriented injection, stimulation, destruction, and guided potential manipulation of neural structures and can be used to create animal models of Parkinson's disease, animal models of epilepsy, tumor model establishment in the brain, learning and memory, intracerebral neural stem cell transplantation, Cerebral ischemia and other studies.
The SA-100 stereotaxic instrument was developed by our company based on the suggestions of several experts in the field of neuroscience research, and based on a variety of similar products. Operations
1. Positioning operation
Step one:Fixed animals:
1) The animals were anesthetized, and after completion of anesthesia, the incisors were toggle open with forceps and stuck inside the square hole of the incisor clamp from the animal adapter, pressing the incisor clamp cross bar.Incisor clip height was adjusted so that the animal interaural line remained consistent with the stainer ear bar line;
2) Depending on the animal body size, high animal body using pads such as foam or plank on the base of the positioning apparatus to keep the head of the animal at the level of the body and prevent respiratory obstruction of the animal;
- The left ear bar was inserted into the ear canal of the animal, and the left and right ear bars were adjusted to keep the animal's head in the central position of the U-shaped opening, and the left ear bars were fixed first and postfixed so that the animal's head could not shake while adjusting the incisor clip screws (which could not be too tight, otherwise affecting respiration).
Step two:Finding a targeting reference site
The following examples were used to search for the bragma points in rats: the scalp of the animal was cut, the periosteum was removed, and the skull diagram of the 290g rat brain was used as an example, as shown in Figure 1. According to the sutures of the rat brain, we searched for the bregma point (bragma) as the reference point for brain positioning, and moved the three-dimensional operator arms to move the position points according to the size mouse brain stereotaxic atlas, such as holding the positioning needle,Then its tip was the localization point) were aligned to the bragma point, and readings in the X (horizontal), y (anterior posterior), and Z (vertical) axes of a brain stereotaxic instrument were recorded (reading method with the same scale bar), using the bragma point as the reference zero point for the three- dimensional coordinate system.
Step three:Localization of experimental areas of the animal brain
According to the atlas finding the coordinate values (i.e., x, y, Z coordinates) of the experimental area to the bragma point, starting from the bragma Reference zero, moving the corresponding distance of the operation arm to the designated position (the Z arm does not move), using the locating needle to make a positioning mark on the surface of the skull, and using a
cranial bur to make an hole.
Step four:Experiment
Hold the syringe, move the z-arm to the surface of the skull, the vertical zero point of the atlas is the surface of the skull, enter from the rat skull opening, and reach the designated depth according to the numerical value of the atlas for injection.
For example: as shown in Figure 1, figure 2, to perform the drug injection experiment in the LV position of rat brain region, first find the bragma point positioning as the zero point and record the respective operant arm position values, then from that point, the x-axis is shifted to the right by 1.2 mm and the y-axis is shifted to the 0.4 mm position posteriorly (because of the precision of vernier caliper is 0.1 mm, according to rounding),Z-axis downshift by 3.5 mm (the position before z-axis downshift is the calvarial opening point), at which point the needle position is the LV point.
2.Reading method:
When measured by vernier cerebral stereotaxic apparatus, the integer part of the measured value was read from the master scale and the decimal part from the vernier scale.The vernier principle is to use the cutting line separation on the master ruler (short term line distance) and the difference between the lines on the vernier to read out the fractional parts, for example: the line distance on the master ruler is 1 mm, on the vernier there are 10 grids, and its line distance is 0.9 mm. When the zero score lines of the two coincide, if the vernier scale moves 0.1 mm, its first score line coincides with the first score line of the main scale; If the vernier scale moves 0.2 mm, its second score line coincides with the second score line of the main scale. By analogy, a fractional portion of the quantity can be read from where the vernier ruler coincides with the scoring line on the master. The difference of 0.1 mm between the master and vernier caliper distance is the minimum reading value for vernier calipers. When reading, first read the millimeter integer on the ruler, that is, the integer part in millimeters. Then see that the few tick marks on the vernier are aligned with the tick marks on the body, as in case the sixth tick is aligned with the tick marks, the decimal part is 0.6 mm (if there is no exactly aligned line, take the line closest to the alignment for reading).The reading results were:
L = integer part + decimal part
Judging which tick on the vernier is aligned with the scale line, the following method can be used: select the adjacent three lines, such as the line on the left is to the right of the scale line, the line on the right is to the left of the scale line, and the middle of that line can then be considered to be aligned.
