General Information
Catalog number: AG00-3367
Synonyms: Chondroitinase, chondroitin ABC lyase, chondroitin ABC eliminase
Gene Source: Proteus vulgaris
Express System: Escherichia coli
EC Number: 4.2.2.20
CAS Number: 9024-13-9
Catalyzed Reactions: Chondroitinase ABC cleaves, via and elimination mechanism, sulfated and non-sulfated polysaccharide chains containing 1-4 linkages between hexosamines and glucuronic acid residues. The reaction yields oligosaccharide products (mainly disaccharides) containing unsaturated uronic acids which can be detected by UV spectroscopty at 232 nm. The enzyme is active on chondroitin sulfates A and C, chondroitin sulfates B (dermatan sulfate), chondroitin and hyaluronic acid.
Substrate Specificity: Chondroitin sulfates A and C, chondroitin sulfates B (dermatan sulfate), chondroitin and hyaluronic acid.
Properties
Molecular weight: 117.3 kDa
pH optimum for activity: 8.0
Optimal temperature range: 37 °C
Purity: ≥ 99% by reversed phase HPLC analysis.
Specific Activity: ≥ 100 IU/mg (substrate: chondroitin sulfate A).
Standard: 1 IU and 5 IU.
Unit Definition: One international unit (IU) of chondroitinase ABC is defined as the amount of enzyme that will liberate 1.0 μmole unsaturated oligosaccharides from chondroitin sulfates A per minute at 30 °C and pH 8.0.
Stability: Expiration is at least 12 months from manufacturing date, kept at -20 °C
