Amyloid beta (Aβ or Abeta) denotes peptides of 36–43 amino acids that are crucially involved in Alzheimer's disease as the main component of the amyloid plaques found in the brains of Alzheimer patients. The peptides derive from the amyloid precursor protein (APP), which is cleaved by beta secretase and gamma secretase to yield Aβ. Aβ molecules can aggregate to form flexible soluble oligomers which may exist in several forms.
Human Amyloid beta 42 (AB42) ELISA Kit employs a two-site sandwich ELISA to quantitate AB42 in samples. An antibody specific for AB42 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyAB42 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for AB42 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of AB42 bound in the initial step. The color development is stopped and the intensity of the color is measured.
