2,3-diphosphoglycerate (2,3-DPG) is a small molecule found at high concentrations in red blood cells where it binds to and decreases the oxygen affinity of hemoglobin. BPGMencodes a multifunctional enzyme that catalyzes 2,3-DPG synthesis via its synthetase activity, and 2,3-DPG degradation via its phosphatase activity.
Human 2,3-Disphosphoglycerate (2,3-DPG) ELISA Kit employs a two-site sandwich ELISA to quantitate 2,3DPG in samples. An antibody specific for 2,3DPG has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any2,3DPG present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for 2,3DPG is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of 2,3DPG bound in the initial step. The color development is stopped and the intensity of the color is measured.
