NF-kappa-B is a ubiquitous transcription factor involved in several biological processes. It is held in the cytoplasm in an inactive state by specific inhibitors. Upon degradation of the inhibitor, NF-kappa-B moves to the nucleus and activates transcription of specific genes. NF-kappa-B is composed of NFKB1 or NFKB2 bound to either REL, RELA, or RELB. The most abundant form of NF-kappa-B is NFKB1 complexed with the product of RELA. Four transcript variants encoding different isoforms have been found for RELA.
Human Nuclear factor-KB p65 (NF-KB p65) ELISA Kit employs a two-site sandwich ELISA to quantitate RELA in samples. An antibody specific for RELA has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyRELA present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for RELA is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of RELA bound in the initial step. The color development is stopped and the intensity of the color is measured.
