Placental ribonuclease inhibitor (PRI) is a member of a family of proteinaceous cytoplasmic RNase inhibitors that occur in many tissues and bind to both intracellular and extracellular RNases (summarized by Lee et al., 1988 [PubMed 3219362]). In addition to control of intracellular RNases, the inhibitor may have a role in the regulation of angiogenin (MIM 105850). Ribonuclease inhibitor, of 50,000 Da, binds to ribonucleases and holds them in a latent form. Since neutral and alkaline ribonucleases probably play a critical role in the turnover of RNA in eukaryotic cells, RNH may be essential for control of mRNA turnover; the interaction of eukaryotic cells with ribonuclease may be reversible in vivo.
Human Ribonuclease inhibitor (RNH1/PRI/RNH) ELISA Kit employs a two-site sandwich ELISA to quantitate RNH1 in samples. An antibody specific for RNH1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyRNH1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for RNH1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of RNH1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
