CA153 encodes a membrane-bound protein that is a member of the mucin family. Mucins are O-glycosylated proteins that play an essential role in forming protective mucous barriers on epithelial surfaces. These proteins also play a role in intracellular signaling. This protein is expressed on the apical surface of epithelial cells that line the mucosal surfaces of many different tissues including lung, breast stomach and pancreas. This protein is proteolytically cleaved into alpha and beta subunits that form a heterodimeric complex. The N-terminal alpha subunit functions in cell-adhesion and the C-terminal beta subunit is involved in cell signaling. Overexpression, aberrant intracellular localization, and changes in glycosylation of this protein have been associated with carcinomas. CA153 is known to contain a highly polymorphic variable number tandem repeats (VNTR) domain. Alternate splicing results in multiple transcript variants.
Human Carbohydrate antigen 153 (CA153) ELISA Kit employs a two-site sandwich ELISA to quantitate CA153 in samples. An antibody specific for CA153 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyCA153 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CA153 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CA153 bound in the initial step. The color development is stopped and the intensity of the color is measured.
