Identified in a human 8p amplicon, MFHAS1 is a potential oncogene whose expression is enhanced in some malignant fibrous histiocytomas (MFH). The primary structure of its product includes an ATP/GTP-binding site, three leucine zipper domains, and a leucine-rich tandem repeat, which are structural or functional elements for interactions among proteins related to the cell cycle, and which suggest that overexpression might be oncogenic with respect to MFH.
Human Malignant fibrous histiocytoma-amplified sequence 1 (MFHAS1) ELISA Kit employs a two-site sandwich ELISA to quantitate MFHAS1 in samples. An antibody specific for MFHAS1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMFHAS1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MFHAS1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MFHAS1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
