Phosphatidylinositide 3-kinase-derived membrane-anchored phosphatidylinositides, such as phosphatidylinositol 3-phosphate (PtdIns(3)P), regulate diverse cellular processes. Myotubularin-related protein 12 encoded by MTMR12 functions as an adaptor subunit in a complex with an active PtdIns(3)P 3-phosphatase. Alternatively spliced transcript variants encoding different isoforms have been found for MTMR12.
Human Myotubularin-related protein 12 (MTMR12) ELISA Kit employs a two-site sandwich ELISA to quantitate MTMR12 in samples. An antibody specific for MTMR12 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMTMR12 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MTMR12 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MTMR12 bound in the initial step. The color development is stopped and the intensity of the color is measured.
