Synaptotagmin-15 (Syt15) belongs to the synaptotagmin family, which is a group of membrane-trafficking proteins that contain two C-terminal C2 domains (known as C2A and C2B domains). Most of the synaptotagmins have a unique N-terminal domain that is involved in membrane anchoring or specific ligand binding. Mammals contain 16 synaptotagmins; eight of these bind Ca2+ via their C2-domains (Syt1-Syt3, Syt5-7, Syt9, and Syt10), whereas the other eight, including Syt15, do not.
Human Synaptotagmin-15 (SYT15) ELISA Kit employs a two-site sandwich ELISA to quantitate SYT15 in samples. An antibody specific for SYT15 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anySYT15 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for SYT15 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SYT15 bound in the initial step. The color development is stopped and the intensity of the color is measured.
