SYKis a member of the Syk family of tyrosine kinases. While Syk and Zap-70 are primarily expressed in hematopoietic tissues, there is expression of Syk in a variety of tissues. Within B and T cells respectively, Syk and Zap-70 transmit signals from the B-Cell receptor and T-Cell receptor. Syk plays a similar role in transmitting signals from a variety of cell surface receptors including CD74, Fc Receptor, and integrins. The role of Syk in epithelial malignancies is controversial. Several authors have suggested that abnormal Syk function facilitates transformation in Nasopharyngeal carcinoma and head and neck cancer while other authors have suggested a tumor suppressor role in breast and gastric cancer.
Human Spleen tyrosine kinase (SYK) ELISA Kit employs a two-site sandwich ELISA to quantitate SYK in samples. An antibody specific for SYK has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anySYK present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for SYK is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SYK bound in the initial step. The color development is stopped and the intensity of the color is measured.
