Soluble Receptor Activator of Nuclear Factor-kB (sRANK) is a member of the TNF-receptor superfamily. This receptors can interact with various TRAF family proteins, through which sRANK induces the activation of NF-kappa B and MAPK8/JNK. sRANKr and its ligand are important regulators of the interaction between T cells and dendritic cells. sRANK is also an essential mediator for osteoclast and lymph node development. Mutations at this locus have been associated with familial expansile osteolysis, autosomal recessive osteopetrosis, and Paget disease of bone. Alternatively spliced transcript variants have been described for this locus.
Human Soluble Receptor Activator of Nuclear Factor-kB (sRANK) ELISA Kit employs a two-site sandwich ELISA to quantitate TNFRSF11A in samples. An antibody specific for TNFRSF11A has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTNFRSF11A present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TNFRSF11A is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TNFRSF11A bound in the initial step. The color development is stopped and the intensity of the color is measured.
