Tropomyosin alpha-3 chain is a member of the tropomyosin family of actin-binding proteins involved in the contractile system of striated and smooth muscles and the cytoskeleton of non-muscle cells. Tropomyosins are dimers of coiled-coil proteins that polymerize end-to-end along the major groove in most actin filaments. They provide stability to the filaments and regulate access of other actin-binding proteins. In muscle cells, they regulate muscle contraction by controlling the binding of myosin heads to the actin filament. Mutations in this gene result in autosomal dominant nemaline myopathy, and oncogenes formed by chromosomal translocations involving this locus are associated with cancer. Multiple transcript variants encoding different isoforms have been found for this gene.
Human Tropomyosin alpha-3 chain (TPM3) ELISA Kit employs a two-site sandwich ELISA to quantitate TPM3 in samples. An antibody specific for TPM3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTPM3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TPM3 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TPM3 bound in the initial step. The color development is stopped and the intensity of the color is measured.
