HIP2 belongs to the ubiquitin-conjugating enzyme family. It binds selectively to a large region at the N terminus of huntingtin. This interaction is not influenced by the length of the huntingtin polyglutamine tract. This protein has been implicated in the degradation of huntingtin and suppression of apoptosis.The 200-amino acid HIP2 protein sequence was identical to that of bovine ubiquitin-conjugating enzyme E2-25K. Assays of the interaction of HIP2 with huntingtin revealed that HIP2 binds selectively to a large region at the N terminus of huntingtin. This interaction is not influenced by the length of the huntingtin polyglutamine tract. Western blot analysis detected protein bands of 25, 28, and 45 kD; the 28-kD band appeared to be selective for tissues of the central nervous system.
Human Ubiquitin-conjugating enzyme E2 K (UBE2K) ELISA Kit employs a two-site sandwich ELISA to quantitate UBE2K in samples. An antibody specific for UBE2K has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyUBE2K present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for UBE2K is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of UBE2K bound in the initial step. The color development is stopped and the intensity of the color is measured.
