Ubiquilin-2 is a ubiquitin-like protein (ubiquilin) that shares high degree of similarity with related products in yeast, rat and frog. Ubiquilins contain a N-terminal ubiquitin-like domain and a C-terminal ubiquitin-associated domain. They physically associate with both proteasomes and ubiquitin ligases, and thus thought to functionally link the ubiquitination machinery to the proteasome to affect in vivo protein degradation. This ubiquilin has also been shown to bind the ATPase domain of the Hsp70-like Stch protein. The predicted 624-amino acid PLIC2 protein, also called UBQLN2, shares 72% amino acid identity with PLIC1. Two motifs are conserved in the mammalian PLICs and yeast Dsk2, an N-terminal ubiquitin-like (UBL) domain and a C-terminal ubiquitin-associated (UBA) domain.
Human Ubiquilin-2 (UBQLN2) ELISA Kit employs a two-site sandwich ELISA to quantitate UBQLN2 in samples. An antibody specific for UBQLN2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyUBQLN2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for UBQLN2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of UBQLN2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
