Reduced expression of p600 with interfering RNA abrogated integrin-mediated ruffled membrane formation and prevented activation of integrin-mediated survival pathways. The deduced 5,183-amino acid protein has a calculated molecular mass of 573.5 kD. Immunostaining of human foreskin fibroblasts localized p600 in a continuous meshwork pattern from the nucleus to the cytoplasm, passing through the nuclear envelope. In the cytoplasm, p600 was concentrated at the leading edge of membrane structures.
Human E3 ubiquitin-protein ligase UBR4 (UBR4) ELISA Kit employs a two-site sandwich ELISA to quantitate UBR4 in samples. An antibody specific for UBR4 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyUBR4 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for UBR4 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of UBR4 bound in the initial step. The color development is stopped and the intensity of the color is measured.
