VSIG2 gene contains 7 conserved exons, with each Ig domain encoded by 2 half-domain exons with unusual splicing junctions.The deduced 325-amino acid CTH protein, which is 29% similar to GPA33, contains a signal peptide, an extracellular V-type Ig-like domain followed by a C2-type Ig-like domain, a transmembrane region devoid of charged residues, and a cytoplasmic tail. There are multiple N-glycosylation sites in the extracellular region and phosphorylation sites in the cytoplasmic tail. Northern blot analysis revealed CTH expression in stomach, colon, prostate, trachea, and thyroid gland, with lower levels in bladder and lung. Human V-set and immunoglobulin domain-containing protein 2 (VSIG2) ELISA Kit employs a two-site sandwich ELISA to quantitate VSIG2 in samples. An antibody specific for VSIG2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyVSIG2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for VSIG2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of VSIG2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
