The 70 kilodalton heat shock proteins (Hsp70s or DnaK) are a family of conserved ubiquitously expressed heat shock proteins. Proteins with similar structure exist in virtually all living organisms. The Hsp70s are an important part of the cell's machinery for protein folding, and help to protect cells from stress.
Fish Heat Shock Protein 70 (HSP-70) ELISA Kit employs a two-site sandwich ELISA to quantitate HSP-70 in samples. An antibody specific for HSP-70 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyHSP-70 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for HSP-70 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of HSP-70 bound in the initial step. The color development is stopped and the intensity of the color is measured.
