Milk fat globule-EGF factor 8 protein (MFGE8) gene encodes a preproprotein that is proteolytically processed to form multiple protein products. The major encoded protein product, lactadherin, is a membrane glycoprotein that promotes phagocytosis of apoptotic cells. This protein has also been implicated in wound healing, autoimmune disease, and cancer. Lactadherin can be further processed to form a smaller cleavage product, medin, which comprises the major protein component of aortic medial amyloid (AMA).
Bovine Milk fat globule-EGF factor 8 protein (MFGE8) ELISA Kit employs a two-site sandwich ELISA to quantitate MFGE8 in samples. An antibody specific for MFGE8 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMFGE8 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MFGE8 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MFGE8 bound in the initial step. The color development is stopped and the intensity of the color is measured.
