Matrix Gla protein (MGP) gene encodes a member of the osteocalcin/matrix Gla family of proteins. The encoded vitamin K-dependent protein is secreted by chondrocytes and vascular smooth muscle cells, and functions as a physiological inhibitor of ectopic tissue calcification. Carboxylation status of the encoded protein is associated with calcification of the vasculature in human patients with cardiovascular disease and calcification of the synovial membranes in osteoarthritis patients. Mutations in this gene cause Keutel syndrome in human patients, which is characterized by abnormal cartilage calcification, peripheral pulmonary stenosis and facial hypoplasia.
Bovine Matrix Gla protein (MGP) ELISA Kit employs a two-site sandwich ELISA to quantitate MGP in samples. An antibody specific for MGP has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMGP present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MGP is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MGP bound in the initial step. The color development is stopped and the intensity of the color is measured.
