N-acetyl-D-glucosamine kinase (NAGK) converts endogenous N-acetylglucosamine (GlcNAc), a major component of complex carbohydrates, from lysosomal degradation or nutritional sources into GlcNAc 6-phosphate. Involved in the N-glycolylneuraminic acid (Neu5Gc) degradation pathway: although human is not able to catalyze formation of Neu5Gc due to the inactive CMAHP enzyme, Neu5Gc is present in food and must be degraded. Also has ManNAc kinase activity.
Bovine N-acetyl-D-glucosamine kinase (NAGK) ELISA Kit employs a two-site sandwich ELISA to quantitate NAGK in samples. An antibody specific for NAGK has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyNAGK present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for NAGK is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of NAGK bound in the initial step. The color development is stopped and the intensity of the color is measured.
