The thyroid hormones, triiodothyronine (T3) and its prohormone, thyroxine (T4), are tyrosine-based hormones produced by the thyroid gland that are primarily responsible for regulation of metabolism. T3 and T4 are partially composed of iodine (see molecular model). A deficiency of iodine leads to decreased production of T3 and T4, enlarges the thyroid tissue and will cause the disease known as simple goitre. The major form of thyroid hormone in the blood is thyroxine (T4), which has a longer half-life than T3.
Bovine Thyroxine (T4) ELISA Kit employs a two-site sandwich ELISA to quantitate T4 in samples. An antibody specific for T4 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyT4 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for T4 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of T4 bound in the initial step. The color development is stopped and the intensity of the color is measured.
