Tmub1 encodes for a protein, HOPS/DULP, containing transmembrane domains and a ubiquitin-like domain (UBL) Ubiquitin can be employed to modify proteins by either single or multiple ubiquitin conjugation to mark them for proteasomal destruction, endocytosis and binding to other proteins. Tmub1/HOPS was reported to facilitate the recycling of AMPA receptors into synaptic membrane in cultured primary neurons. Tmub1/HOPS was also found overexpressed during liver regeneration and is an essential constituent of centrosome assembly during cell cycles in culture. Like many UBL domain proteins, the physiological and molecular function of Tmub1 is far from being clear. The resulting mice Tmub1were put through an extensive battery of assays to discover physiological and behavioral phenotypes.
Bovine Transmembrane and ubiquitin-like domain-containing protein 1 (TMUB1) ELISA Kit employs a two-site sandwich ELISA to quantitate TMUB1 in samples. An antibody specific for TMUB1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTMUB1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TMUB1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TMUB1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
