Troponin is a component of thin filaments (along with actin and tropomyosin), and is the protein complex to which calcium binds to trigger the production of muscular force. Troponin itself has three subunits, TnC, TnI, and TnT, each playing a role in force regulation. TnT is a tropomyosin-binding subunit which regulates the interaction of troponin complex with thin filaments. cTnT is probably less cardiac specific. Expression of cTnT in skeletal tissue of patients with chronic skeletal muscle injuries has been described.
Bovine Cardiac Troponin T (cTn-T) ELISA Kit employs a two-site sandwich ELISA to quantitate TNNC1 in samples. An antibody specific for TNNC1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTNNC1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TNNC1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TNNC1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
