The advanced slide surface treatment technology (TC treatment → super adsorption) can promote the cell to adhere to the wall and grow on the slide, and the cell adheres to the wall firmly.
Even in the later stage of immunohistochemistry, immunofluorescence and in situ hybridization, it is not easy to take off the slide, so as to avoid the damage of traditional methods from the culture bottle to the slide. The use of one-step cell climbing tablets does not need pretreatment, so as to avoid the problems of cell peeling in the later treatment.

Precautions for cell climbing operation:
1. In the super clean table, use 75% ethanol to wipe the outer packing aluminum foil bag
2. Disassemble the aluminum foil bag, take out the built-in utensils, and use the tip tweezers to take out the built-in climbing piece and put it into the used culture plate or culture dish,
3. Digest the cells with trypsin and blow them to form single cell suspension (Note: This is very important, which is related to the quality of the films that will climb out in the future)
4. When planting cells, according to the size of the slide, first drop a small amount of culture medium at the position where the slide is to be placed in each hole. The purpose is to make the slide and the culture dish adhere to the tension of the culture medium, and then put the slide to prevent the slide from floating when the cell suspension is added, resulting in double-layer cell sticking. Pay attention to aseptic operation in the whole process.
5. Select the appropriate cell density to plant in the culture plate according to your own needs

When using the cell climbing sheet (for example, put the climbing sheet into the 6-hole plate clock, the area of the hole bottom of the 6-hole plate is often larger than that of the climbing sheet, when adding the cell suspension, it is often that the cell spreads over the whole hole bottom, sometimes, the phenomenon of cell growth edge gathering is that the cell density near the wall edge is higher, so the number of cells on the central cell climbing sheet is relatively small), which is better to do The method is that after putting the climbing tablet into the hole, when adding the cell suspension, only drop it onto the climbing tablet until the liquid is full of the whole climbing tablet, but it is not easy to overflow the edge of the climbing tablet, put it into the incubator, take it out after the cell adheres to the wall, then drop the culture medium to cover the whole hole bottom, and continue to culture, so that the cell growth density on the climbing tablet will be very concentrated.
7. Put the cell on the climbing tablet, and then add any reagent. Pour a little liquid along the wall of the culture plate until the cell is submerged by the liquid. Try not to flush the cell outside the climbing tablet!
8. Take the slide after acting for a certain time. When taking the slide, due to the tight combination of the slide and the bottom of the culture dish, the tension is large. Generally, make a small hook from the tip of the syringe needle to the back, so that the slide can be gently lifted up and taken out with small tweezers. If you want to do experiments at such time points as 24h, 48h, 72h, etc., when you take out the required number of climbing pieces, use an alcohol lamp flame to burn the needles and tweezers. The last one can be further cultured.
9. Cell slides, with multi-layer packaging, can be unpacked in the sterile state of super clean platform, and the unused slides can be repackaged according to the original packaging method
10. There is a positive charge on the surface of the climbing plate, please do not touch it with your hand, otherwise the effect will be poor!
