MUC1 is anchored to the apical surface of many epithelia by a transmembrane domain, with the degree of glycosylation varying with cell type. It also includes a 20 amino acid variable number tandem repeat (VNTR) domain, with the number of repeats varying from 20 to 120 in different individuals.The protein serves a protective function by binding to pathogens and also functions in a cell signaling capacity. Overexpression, aberrant intracellular localization, and changes in glycosylation of this protein have been associated with carcinomas. Multiple alternatively spliced transcript variants that encode different isoforms of this gene have been reported, but the full-length nature of only some has been determined.
Rat Mucin-1 (MUC1) ELISA Kit employs a two-site sandwich ELISA to quantitate MUC1 in samples. An antibody specific for MUC1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMUC1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MUC1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MUC1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
