Carnitine (β-hydroxy-γ-N-trimethylaminobutyric acid, 3-hydroxy-4-N,N,N- trimethylaminobutyrate) is a quaternary ammonium compound involved in metabolism in most mammals, plants and some bacteria. Carnitine may exist in two isomers, labeled D-carnitine and L-carnitine, as they are optically active. At room temperature, pure carnitine is a white powder, and a water-soluble zwitterion with low toxicity. Carnitine only exists in animals as the L-enantiomer, and D-carnitine is toxic because it inhibits the activity of L-carnitine.
Rat Total Carnitine ELISA Kit employs a two-site sandwich ELISA to quantitate Total Carnitine in samples. An antibody specific for Total Carnitine has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTotal Carnitine present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for Total Carnitine is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of Total Carnitine bound in the initial step. The color development is stopped and the intensity of the color is measured.
