Anti-neutrophil cytoplasmic antibodies (ANCAs) are a group of autoantibodies, mainly of the IgG type, against antigens in the cytoplasm of neutrophil granulocytes (the most common type of white blood cell) and monocytes. They are detected as a blood test in a number of autoimmune disorders, but are particularly associated with systemic vasculitis, so called ANCA-associated vasculitides.
Rat Myeloperoxidase-antineutrophil cytoplasmic antibody IgG (MPO-ANCA-IgG) ELISA Kit employs a two-site sandwich ELISA to quantitate MPO-ANCA-IgG in samples. An antibody specific for MPO-ANCA-IgG has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMPO-ANCA-IgG present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MPO-ANCA-IgG is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MPO-ANCA-IgG bound in the initial step. The color development is stopped and the intensity of the color is measured.
