DEK encodes a protein with one SAP domain. This protein binds to cruciform and superhelical DNA and induces positive supercoils into closed circular DNA, and is also involved in splice site selection during mRNA processing. Chromosomal aberrations involving this region, increased expression of DEK, and the presence of antibodies against this protein are all associated with various diseases. Two transcript variants encoding different isoforms have been found for DEK.
Rat Protein DEK (DEK) ELISA Kit employs a two-site sandwich ELISA to quantitate DEK in samples. An antibody specific for DEK has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyDEK present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for DEK is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of DEK bound in the initial step. The color development is stopped and the intensity of the color is measured.
