Intron 1 is approximately 1.7 kb long and is located in the 5-prime untranslated region of the MUC7 cDNA.Intron 2 spans approximately 6 kb and is located close to the boundary of the putative liter peptide and secreted protein. The entire region encoding the secreted peptide is located on exon 3, spanning approximately 2.2 kb.Bobek et al. Mouse Mucin-7 (MUC7) ELISA Kit employs a two-site sandwich ELISA to quantitate MUC7 in samples. An antibody specific for MUC7 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMUC7 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MUC7 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MUC7 bound in the initial step. The color development is stopped and the intensity of the color is measured.
