CYP7A1 encodes a member of the cytochrome P450 superfamily of enzymes. The cytochrome P450 proteins are monooxygenases which catalyze many reactions involved in drug metabolism and synthesis of cholesterol, steroids and other lipids. This endoplasmic reticulum membrane protein catalyzes the first reaction in the cholesterol catabolic pathway in the liver, which converts cholesterol to bile acids. This reaction is the rate limiting step and the major site of regulation of bile acid synthesis, which is the primary mechanism for the removal of cholesterol from the body. Polymorphisms in the promoter of CYP7A1 are associated with defects in bile acid synthesis.
Human Cholesterol 7 alpha-Hydroxylase (CYP7A) ELISA Kit employs a two-site sandwich ELISA to quantitate CYP7A1 in samples. An antibody specific for CYP7A1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyCYP7A1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CYP7A1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CYP7A1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
