CYP8B1 encodes a member of the cytochrome P450 superfamily of enzymes. The cytochrome P450 proteins are monooxygenases which catalyze many reactions involved in drug metabolism and synthesis of cholesterol, steroids and other lipids. This endoplasmic reticulum membrane protein catalyzes the conversion of 7 alpha-hydroxy-4-cholesten-3-one into 7-alpha,12-alpha-dihydroxy-4-cholesten-3-one. The balance between these two steroids determines the relative amounts of cholic acid and chenodeoxycholic acid both of which are secreted in the bile and affect the solubility of cholesterol. CYP8B1 is unique among the cytochrome P450 genes in that it is intronless.

An antibody specific for CYP8B1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyCYP8B1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CYP8B1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CYP8B1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
