Proteins of the matrix metalloproteinase (MMP) family are involved in the breakdown of extracellular matrix in normal physiological processes, such as embryonic development, reproduction, and tissue remodeling, as well as in disease processes, such as arthritis and metastasis. Most MMP's are secreted as inactive proproteins which are activated when cleaved by extracellular proteinases. Matrix metalloproteinase-20 encoded by MMP20 degrades amelogenin, the major protein component of dental enamel matrix, and thus thought to play a role in tooth enamel formation. A mutation in MMP20, which alters the normal splice pattern and results in premature termination of the encoded protein, has been associated with amelogenesis imperfecta. MMP20 is part of a cluster of MMP genes located on chromosome 11q22.3.

Human Matrix metalloproteinase-20 (MMP20) ELISA Kit employs a two-site sandwich ELISA to quantitate MMP20 in samples. An antibody specific for MMP20 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMMP20 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MMP20 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MMP20 bound in the initial step. The color development is stopped and the intensity of the color is measured.
