★ Technical Manual
Human Umbilical Cord Mesenchymal Stem Cell Serum-free Medium (HUMSC-SFM)
★ Catalog NO: BW110022
★ Components
490 ml of Basal Medium
5 ml of Human Umbilical Cord Mesenchymal Stem Cell Supplement (HUMSCS)
5 ml of penicillin/streptomycin solution (P/S)
★ Product Description
Human Umbilical Cord Mesenchymal Stem Cell Serum-free Medium (HUMSC-SFM) contains defined components including essential and non-essential amino acids, vitamins, organic and inorganic compounds, hormones, growth factors, trace minerals and other supplements. The product is suitable for culturing human umbilical cord mesenchymal stem cells. The product is strictly aseptic without any contamination of virus or mycoplasma, and exhibits stable performance. By using this medium, human umbilical cord mesenchymal stem cells can be quickly expanded with minimal differentiation.
★ Instruction for Use
It is strongly recommended that cell plates should be precoated with CELLstart™1:50 (Cat. No. A10142-01, GIBCO) in Dulbecco’s Phosphate Buffered Saline with calcium and magnesium (Cat. No. A12858, GIBCO).
1. Dilute CELLstart™1:50 (Cat. No. A10142-01, GIBCO) in Dulbecco’s Phosphate Buffered Saline with calcium and magnesium (Cat. No. A12858, GIBCO).
2. Add diluted CELLstart™ to culture container at a final volume per surface area of 0.078 mL/cm2. Refer to table for respective culture container:
Culture Container Surface Area (cm2) Volume of 1:50 Diluted CELLstart
6-well plate 9.6 750ul per well
12-well plate 3.2 250ul per well
24-well plate 2.0 160ul per well
3. Incubate in a 37°C, 5% CO2 incubator for 2 hours. It is recommended to coat culture containers the day of use or the day before. If precoating the day before, the culture container must be stored at 2 to 8°C wrapped with Parafilm® to avoid drying.
4. Pipette out diluted CELLstart™ from culture container and discard. Culture vessel is ready for the addition of cells.
*Note: It is not necessary to rinse the culture container after removal of CELLstart™. The bottom of the coated culture container should have a clear and wet appearance.
5. Thaw penicillin/streptomycin solution (P/S) overnight at 2 to 8 ℃.
6. Add the above thawed solutions to the Basal Medium, gently swirl the bottle to ensure the mixture homogeneity.
7. Thaw the Human Umbilical Cord Mensenchymal Stem Cell Supplement (HUMSCS) in water bath (37℃) and then add it into the above mixture to make the complete medium. 8. Store the complete medium in the dark at 4℃.
★ Product Use
This product is for laboratory research use only. It is not approved for human or animal use, or for application in in vitro diagnostic procedures.
★ Storage condition
Basal Medium should be stored at 4℃, while HUMSCS and P/S solution should be stored in the dark at -20℃.

ArkOne Biotechnologies, Inc. is a global distributor of high quality biochemical reagents and lab research tools for protein chemistry, molecular biology, and cell biology including a series of DNA vectors, purified proteins of high quality, and media for cell culture. The pre-selected media are suitable for stem cell culture, especially for those mammalian stem cells isolated from bone marrow, umbilical cord, and adipose tissues. By using these media, cells can be quickly expanded with minimal differentiation. The serum media contain essential and non-essential amino acids, vitamins, organic and inorganic compounds, hormones, growth factors, trace minerals and pre-selected fetal bovine serum and other supplements. The serum-free media contain defined animal-free components and key cytokine supplements, targeting clinical applicaiton. We also provide packaging services for a variety of viruses including intergrative viruses and non-integrative viruses of high titer. Integrative viruses are suitable for rapid generation of stable cell lines. These viruses include retrovirus and classical lentivirus. Non-integrative viruses include modified lentivirus, adenovirus and adeno-associated viruses (AAV) with serotypes ranging from AAV2/1 to AAV2/9. Non-integrative viruses can serve as excellent options for transient transfection as well as for gene delivery treatment (http://).
