Working Principle of Real-time PCRsFluorescence quantitative PCR technique is a kind of analyzing method that to add expert probes marked with fluorescent group into PCR reaction solution, and during the PCR process, owing to the hydrolyzation of the fluorescence probe by Taq polymerase, fluorescence signals will be accumulated so to realize real-time monitoringthe whole PCR course and finally by comparing to the standard curve to realize the quantitative analysis of unknowntemplate. Here, our fluorescence probes adopt the latest technique of Taqman, "TaqmanMGB",
which has two main advantages compared to the traditional Taqman probe: ① Probe 3 × is marked with fluorescent quencher that can't illuminate by itself in stead of common illuminant Tamara fluorescence label. This new technique can reduce the fluorescence background and improve resolving power of fluorescence spectrum, especially improve the resolving power among fluorescence spectrums of different reportersduring SNP analysis.
② Probe 3 × combines Minor groove binder so as to increase the Tm value and strengthen the hybridization stability.
Features of Taqman MGB ● Low fluorescence background and high sensitivity.
● Enlarge the difference of Tm values between target template and various non-specific strips as same as the specificity.
● New fluorescence Taqman MGB probe can improve hybridization stability with higher resolving power compared to traditional Taqman probe.
● Long-life storage.
● Good experimental repetition.
