Description
pUM-T vector is designed for PCR cloning by BioTeke. pUM-T vector derived from pUC18 provides 3’ deoxythymidine (T) by adding certain sequence and specific procedure. Taq polymerase has a non template-dependent terminal transferase activity that adds a single deoxyadenosine (A) to the 3’ ends of PCR products. This allows PCR inserts to ligate efficiently with the vector. The insert does not change lac Z reading frame, so positive clones can be picked by blue/white screening. In Addition, pUM-T vector contains pUC18 multiple cloning sites. The insert can be confirmed by double digestion or sequenced using M13 universal primers.
