Nicotinamide adenine dinucleotide phosphate, abbreviated NADP+ or, in older notation, TPN (triphosphopyridine nucleotide), is a cofactor used in anabolic reactions, such as lipid and nucleic acid synthesis, which require NADPH as a reducing agent. NADPH is the reduced form of NADP+. NADP+ differs from NAD+ in the presence of an additional phosphate group on the 2' position of the ribose ring that carries the adenine moiety.

An antibody specific for NADPH has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyNADPH present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for NADPH is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of NADPH bound in the initial step. The color development is stopped and the intensity of the color is measured.
