NEDD8-activating enzyme E1 regulatory subunit (NAE1) binds to the beta-amyloid precursor protein. Beta-amyloid precursor protein is a cell surface protein with signal-transducing properties, and it is thought to play a role in the pathogenesis of Alzheimer's disease. In addition, NAE1 can form a heterodimer with UBE1C and bind and activate NEDD8, a ubiquitin-like protein. NAE1 is required for cell cycle progression through the S/M checkpoint. Three transcript variants encoding different isoforms have been found for NAE1.

An antibody specific for NAE1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyNAE1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for NAE1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of NAE1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
