Neurocalcin-delta (NCALD) encodes a member of the neuronal calcium sensor (NCS) family of calcium-binding proteins. NCALD contains an N-terminal myristoylation signal and four EF-hand calcium binding loops. NCALD is cytosolic at resting calcium levels; however, elevated intracellular calcium levels induce a conformational change that exposes the myristoyl group, resulting in protein association with membranes and partial co-localization with the perinuclear trans-golgi network. NCALD is thought to be a regulator of G protein-coupled receptor signal transduction. Several alternatively spliced variants of this gene have been determined, all of which encode the same protein; additional variants may exist but their biological validity has not been determined.
Chicken Neurocalcin-delta (NCALD) ELISA Kit employs a two-site sandwich ELISA to quantitate NCALD in samples. An antibody specific for NCALD has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyNCALD present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for NCALD is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of NCALD bound in the initial step. The color development is stopped and the intensity of the color is measured.
