POT1 is a member of the telombin family and encodes a nuclear protein involved in telomere maintenance. Specifically, this protein functions as a member of a multi-protein complex that binds to the TTAGGG repeats of telomeres, regulating telomere length and protecting chromosome ends from illegitimate recombination, catastrophic chromosome instability, and abnormal chromosome segregation. Increased transcriptional expression of this gene is associated with stomach carcinogenesis and its progression. Alternatively spliced transcript variants have been described.Human POT1 encodes a 71-kD protein whose N-terminal region shares 26% identity with the N terminus of S. pombe Pot1. POT1 is a housekeeping gene required to ensure the integrity of chromosome ends in all cells.
Chicken Protection of telomeres protein 1 (POT1) ELISA Kit employs a two-site sandwich ELISA to quantitate POT1 in samples. An antibody specific for POT1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPOT1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for POT1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of POT1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
