Deoxyhypusine synthase (DHPS) gene encodes a protein that is required for the formation of hypusine, a unique amino acid formed by the posttranslational modification of only one protein, eukaryotic translation initiation factor 5A. The encoded protein catalyzes the first step in hypusine formation by transferring the butylamine moiety of spermidine to a specific lysine residue of the eukaryotic translation initiation factor 5A precursor, forming an intermediate deoxyhypusine residue.
Bovine Deoxyhypusine synthase (DHPS) ELISA Kit employs a two-site sandwich ELISA to quantitate DHPS in samples. An antibody specific for DHPS has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyDHPS present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for DHPS is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of DHPS bound in the initial step. The color development is stopped and the intensity of the color is measured.
