Dihydrotestosterone (DHT), or 5α-dihydrotestosterone (5α-DHT), also known as 5α-androstan-17β-ol-3-one, is an endogenous androgen sex steroid and hormone. The enzyme 5α-reductase catalyzes the formation of DHT from testosterone in certain tissues including the prostate gland, seminal vesicles, epididymides, skin, hair follicles, liver, and brain. This enzyme mediates reduction of the C4-5 double bond of testosterone. Relative to testosterone, DHT is considerably more potent as an agonist of the androgen receptor (AR).
Bovine Dihydrotestosterone (DHT) ELISA Kit employs a two-site sandwich ELISA to quantitate DHT in samples. An antibody specific for DHT has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyDHT present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for DHT is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of DHT bound in the initial step. The color development is stopped and the intensity of the color is measured.
