Malate dehydrogenase catalyzes the reversible oxidation of malate to oxaloacetate, utilizing the NAD/NADH cofactor system in the citric acid cycle. The protein encoded by this gene is localized to the mitochondria and may play pivotal roles in the malate-aspartate shuttle that operates in the metabolic coordination between cytosol and mitochondria. Malate dehydrogenase, mitochondrial (MDH2) protein includesd in Metabolism and Citrate cycle (TCA cycle) pathway.This protein acts as an enzyme. It is known to catalyze the following reaction : (S)-malate + NAD(+) = oxaloacetate + NADH , and the Enzyme activity is enhanced by acetylation.
Bovine Malate dehydrogenase, mitochondrial (MDH2) ELISA Kit employs a two-site sandwich ELISA to quantitate MDH2 in samples. An antibody specific for MDH2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMDH2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MDH2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MDH2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
