The Overture™ Robotic Peptide Library Synthesizer was designed to generate and synthesize peptide libraries in an easy to use, high-throughput format.  The Overture's™ flexible reaction vessel configurations and software allow libraries from 4 to 96 compounds to be made, including overlapping peptides for epitope mapping, alanine-scanning libraries, positional scanning libraries, random libraries, truncation peptide libraries and scrambled peptide libraries for sequence optimization.  The Overture's™ flexible, easy to use software features easy sequence importing, automatic sequence placement, automatic library generation, automated cleaning routines and report and log file generation.  It is also the only robotic peptide synthesizer to feature on-board automated cleavage without any user intervention!

Special features include the Safe-Response feature which automatically drains and rinses the resin with solvent when an error occurs, as well as E-mail Notification which can send an email to a computer or cell phone with the status of a synthesis, for worry-free operation.  Other features include a workstation utility, which allows syntheses to be created on a desktop computer, and then transferred to the Overture™ with a USB stick!  A unique door-opening mechanism and convenient solvent cabinet save bench space.

    Features

    6 Independent Reaction Vessel Blocks
The Overture™ features 6 independent reaction vessel blocks available in 2 configurations which hold 16 (1.3 mL, 10 mL) RV's or 4 (1.3 mL, 10 mL, 40 mL, 45 mL) RV's.   Each reaction vessel block is independent, so you can run 6 different programs simultaneously on the instrument, and use any configuration for any RV block.  Run anywhere from 4 to 96 peptides at the same time!

    49 Amino Acid Positions
The Overture™ can hold up to 49 individual amino acid dispensers capable of infinitely variable delivery volumes accurate to 50 uL.  Each dispenser is primed automatically by the synthesizer.  No rinsing is required between amino acid deliveries, saving solvent and time!

    Full-Color Chemical-Resistant Touchscreen
Software operation is easy with this feature, and with no external computer, the Overture™ saves you valuable lab space as well!

    Flexible Fluid Delivery
Select from 1-channel or 4-channel wands to dispense reagents, or use fast flood-fill deliveries for washing!

    Variable
    Vortex Mixing
The Overture™ features continuous variable vortex mixing of the reaction block during syntheses, so every reaction vessel gets the mixing intensity it needs regardless of scale or protocol.

    Single-Point Calibration
Calibrate the robotic arm in less than 5 minutes with automated single-point calibration!

    Automatic Library Generation
Create your own libraries on the Overture™ software!  Choose from overlapping peptide libraries for epitope mapping to alanine-scanning libraries, positional scanning libraries, random libraries, truncation peptide libraries and scrambled peptide libraries.  The Overture™ will automatically assign reaction vessel positions for each peptide in a library.

    User Logging
The Overture™ provides different levels of security for different users with its login screen.
System access limited to authorized individuals
User ID and password provide an electronic signature for each user
Users may login as an Administrator, Designer or Runner

    Personalized User Databases
Individual users can create their own programs and peptide sequences that can only be accessed when they are logged in. Files can be created on the Overture™ or on an external computer and transferred to the Overture™ via a portable USB drive:

    E-mail Notification Feature
Have the Overture™ send an email to your computer or cell phone at a specified program step, at the beginning of each cycle, or when an error occurs.

    Safe Response Feature
The Overture™ can rinse and drain your reaction with a solvent of your choice if your synthesis is interrupted for any reason.

    Automated Cleavage
The Overture™ is the only robotic synthesizer that can cleave your peptides on the instrument without user intervention!  Deliver TFA to the reaction vessel from a solvent bottle or an amino acid dispenser.  Afterwards, collect your cleaved peptides in 12 or 50 mL centrifuge vials located directly below the reaction vessel blocks, in convenient centrifuge-compatible holders!  Add ether directly to the collection vials and centrifuge to precipitate your peptide!

Protein Technologies, Inc. (PTI) was founded in 1985 by researchers affiliated with the University of Arizona in Tucson. In 1990, we launched our first peptide synthesizer, the PS3™, and since then we have manufactured and sold the world's finest solid phase synthesizers.

The PS3™ peptide synthesizer met with immediate success and continues to be one of the most reliable, cost effective, and easy to use instruments on the market today.

In 1993 we introduced the Symphony®, a fully automated peptide synthesizer featuring 12 independent reaction vessels that has dominated the multi-channel solid-phase synthesis market. The Symphony® is ideal for custom peptide synthesis because individual reaction vessels may be started, paused, or stopped regardless of what the other reaction vessels are doing, making it equivalent to 12 separate synthesizers in one! The Symphony® is widely regarded as perhaps the finest synthesizer available, and there are hundreds of Symphonys®operating worldwide.

In 1996 we introduced the Sonata®, a large scale automated peptide synthesizer. We included the well-established fluid movement of the Symphony®, as well as its ease of use, reagent optimization, and automated cleavage. We also added a mechanical agitator to assist nitrogen bubbling in order to efficiently mix large amounts of resin in the larger reaction vessels. The Sonata® can produce from grams to near-kilograms of expected product. In 2009, we introduced the Sonata® XT which doubles the capacity of the original Sonata® peptide synthesizer!

In 2000 we introduced the Quartet™, a 4 channel version of the Symphony® peptide synthesizer. It has all of the advantages of the Symphony® and it can be upgraded to the full Symphony® if the user's needs should grow.

In 2006, we introduced the Prelude®, a 6-channel parallel peptide synthesizer that features no-prime deliveries and extra amino acid bottles for expensive monomer additions. It can also rinse your reaction when an error occurs and send an E-mail status report to your computer or cell phone.

In 2007, we introduced the Tribute®, a revolutionary 2-channel parallel peptide synthesizer featuring a full-color chemical-resistant touchscreen, a 101-position amino acid vial rack, amino acid barcode scanning, fully-adjustable vortex and nitrogen mixing, as well as individual user logging. It also features E-mail notification and washing on error like the Prelude®. In 2009, we added the unique IntelliSynth™ UV-Monitoring and Feedback Control System to the Tribute® peptide synthesizer, which is the only UV-Monitoring system that monitors during the deprotection reaction, so it can control both the deprotection time as well as the number of repeats. In 2012, we added infrared (IR) heating to the Tribute®, giving it the fastest time to temperature of any heating technology on the market today! In addition, unlike other rapid heating methods, infrared heating can be performed on more than one reaction vessel at once, and is compatible with vortex mixing, so you can be sure you have an even temperature profile throughout your reaction vessel. Other heated systems that rely on nitrogen mixing may have hot and cold spots, which could compromise the consistency of your chemistry.

In 2010, we introduced the Overture™ robotic peptide library synthesizer. The Overture™ is the most flexible robotic platform on the market today. In addition to powerful library generation tools, the Overture™ is the only robotic platform that supports multiple, simultaneous protocols, the synthesis of different scales at the same time, and on-board automated cleavage without user intervention!

In 2012, we introduced the Symphony® X. With twice the number of reaction vessels as the original Symphony®, the Symphony® X can synthesize up to 12 peptides using preactivation or up to 24 peptides using in situ activation! It also features up to 40 amino acid positions, no-prime deliveries for expensive reagent additions, infrared heating, UV-monitoring, E-mail notification and more! With its 12 independent reactor pairs, the Symphony® X is the perfect instrument for peptide production or protocol optimization.

We have built our company on the belief that our products must be the highest possible quality, and we support our products with a dedicated field service team. We are proud of our reputation for reliability and value the trust our customers and partners have placed in PTI. Today we continue to grow and innovate to serve the needs of the peptide synthesis community.
